It recognizes xenobiotic compounds including steroid hormones and activates the transcription of a variety of genes that are involved in the metabolism of steroid hormones. The human PXR (pregnane X receptor) is expressed in adrenal, testis, and ovary, and it is the first known member of the nuclear receptor superfamily that regulates both the expression of a variety of genes encoding drug metabolizing enzymes and steroid hormone synthesis. The expression of human PXR in adrenocortical H295R cells has been examined, and it was found to be a functional glucocorticoid receptor. The glucocorticoid agonist, dexamethasone, stimulates the PXR-dependent transcription of CYP3A4, the gene encoding P-glycoprotein. This action of dexamethasone was blocked by the glucocorticoid receptor antagonist, RU486.
